اثر عصارة سیر بر رشد، تغذیه و ترکیب شیمیایی لاشة کفال ماهی (Mugil cephalus)
در این تحقیق، اثر عصارة سیر بر شاخصهای رشد (افزایش وزن بدن، نرخ رشد ویژه، فاکتور وضعیت، طول نهایی و وزن نهایی)، تغذیه (ضریب تبدیل غذا، راندمان مصرف پروتئین و راندمان تولید پروتئین) و ترکیب بدن (میزان پروتئین، چربی، رطوبت و خاکستر) در لارو کفالماهیان در 4 تیمار مختلف (سه تکرار در هر تیمار) به مدت 30 روز بررسی شد. به این منظور، لارو کفالماهیان با میانگین وزنی g 01/0±45/0 با جیرة غذایی حاوی 5/0، 1 و 3 درصد عصارة سیر تغذیه شدند و تیمار شاهد با جیرة غذایی فاقد عصارة سیر تغذیه شد. نتایج نشان داد که در پایان آزمایش، بالاترین وزن نهایی g 05/0±78/0، طول نهایی cm 14/0±04/4، بیشترین میزان افزایش وزن % 19/17±13/85، نرخ رشد ویژه % 31/0±93/1، بالاترین نرخ تولید پروتئین % 58/67±05/415، کمترین میزان چربی لاشه % 01/0±37/3 و بیشترین میزان پروتئین لاشه % 03/0±99/20 در تیمار حاوی 3 درصد عصارة سیر مشاهده شد که با تیمار شاهد تفاوت معنیداری داشت (05/0>P). در مجموع، بر اساس نتایج این تحقیق، افزودن 3 درصد عصارة سیر به جیرة غذایی لارو کفالماهیان به منظور بهبود شاخصهای رشد، تغذیه و کیفیت لاشه در این ماهیان پیشنهاد میشود.

تأثیر جایگزینی روغن ماهی با روغنهای گیاهی در توسعة گناد و تغییرات سطح برخی هورمونهای جنسی ماهی قزلآلای رنگینکمان ماده (Oncorhynchus mykiss)
چکیده
به منظور بررسی تأثیر جیرههای غذایی حاوی روغنهای مختلف گیاهی در رشد تخمدان و میزان ترشح هورمونهای جنسی در ماهی قزلآلای رنگینکمان (Oncorhynchus mykiss)، چهار جیرة آزمایشی بر پایة 15% چربی فرموله شد. در تیمار شاهد 100% روغن مورد استفاده روغن ماهی بود و در تیمارهای اول، دوم و سوم از 25% روغن ماهی و به ترتیب 75% روغن سویا، کانولا و کتان استفاده شد. آزمایش به صورت طرح کاملاً تصادفی در چهار تیمار و سه تکرار طراحی شد. پس از شش هفته تغذیة ماهیان با وزن متوسط 5/2 ±200 گرم، بافت تخمدان و میزان هورمونهای استروئیدی (تستسترون، 17-بتا استرادیول و پروژسترون) ارزیابی شدند. میزان توسعة بافت تخمدان همچنین میزان هورمونهای تستسترون و 17-بتا استرادیول در تیمار اول به طور معنیداری بیشتر از سایر تیمارها بود (05/0 > P)، در حالی که میزان هورمون پروژسترون در بین تمام تیمارها و تیمار شاهد اختلاف معنیداری را نشان نداد. با توجه به مطالب ارائهشده، روغن سویا بهمنزلة بهترین روغن در جیرة غذایی ماهی قزلآلای رنگینکمان برای توسعة بافت تخمدان و افزایش میزان هورمونهای جنسی پیشنهاد میشود.

بررسی تأثیر ویتامین C جیرة غذایی در رشد، ترکیب شیمیایی بدن و فعالیت برخی آنزیمهای آنتیاکسیدانی بچهماهی نورس آزاد دریای خزر (Salmo trutta caspius
به منظور بررسی عملکرد ویتامین C در ترکیب شیمیایی بدن و فعالیت آنزیمهای آنتیاکسیدانی شامل سوپراکسید دیسموتاز و کاتالاز در بچهماهی نورس آزاد دریای خزر (Salmo trutta caspius)، پنج جیرة غذایی حاوی سطوح مختلف ویتامین C (صفر (شاهد)، 50، 250، 750 و 1500 میلیگرم ویتامین به ازای هر کیلوگرم غذا) در سه تکرار و به مدت شش هفته به بچهماهیان با میانگین وزنی 02/0± 1/0 گرم خورانده شد. پس از گذشت شش هفته پرورش، نتایج شاخصهای رشد و زندهمانی اختلاف معنیداری در بین تیمارها نشان داد (05/0>P) پایینترین درصد میانگین افزایش وزن (BWG)، نرخ رشد ویژه (SGR) و زندهمانی در گروه شاهد و بالاترین درصد این فاکتورها در گروه تغذیهشده با سطح mg/kg 750 مشاهده شد. میزان تجمع ویتامین C در بافت با افزایش آن در جیرة غذایی رابطة مثبتی نشان داد. نیاز ویتامین C بر اساس درصد افزایش وزن بدن mg/kg 1/569 تخمین زده شد. نتایج بررسی ترکیب شیمیایی بدن نشان داد که مقادیر رطوبت، خاکستر، چربی و پروتئین اختلاف معنیداری بین تیمارها ندارند (05/0 <P). تأثیر تست استرس محدودیت اکسیژنی در فعالیت آنزیمهای آنتیاکسیدانی سوپراکسید دیسموتاز و کاتالاز نشان داد که فعالیت این آنزیمها بعد از اعمال تست استرس نسبت به قبل از آن افزایش معنیداری در تیمار کنترل داشته است (05/0>P) در حالی که فعالیت آنزیمی سایر تیمارها پس از اعمال استرس اختلاف معنیداری نسبت به قبل از استرس نشان نداد (05/0<P). بر اساس نتایج، سطح mg/kg 750 ویتامین C برای رشد و زندهمانی بچهماهیان نورس آزاد دریای خزر پیشنهاد میشود.

تأثیر دورههای نوری و شدت نور مختلف بر قابلیت تولید تخم، درصد تخمگشایی، زمان رسیدگی جنسی و نسبت جنسی در Acartia clausi
هدف از این تحقیق بررسی تأثیر دورهها و شدت نور متفاوت در قابلیت تولید تخم، درصد تخمگشایی، زمان رسیدگی جنسی و نسبت جنسی بالغان در گونةAcartia clausi در شرایط آزمایشگاهی بوده است. این زئوپلانکتوناز سواحل استان مازندران به وسیلة تور پلانکتونگیر 100 میکرونی جمعآوری شد و آزمایشها در پنج تیمار 24:00، 18:06، 12:12، 06:18، 00:24 روشنایی/ تاریکی و در دو شدت نوری 50 و 1000 لوکس انجام شدند. نتایج نشان داد که بیشترین میزان تولید تخم در شدت 50 لوکس و در 18 ساعت روشنایی، به میزان 31/4±02/20 عدد و کمترین آن در شدت 1000 لوکس و 24 ساعت تاریکی به میزان 15/2±15/8 عدد بوده است. بیشترین درصد تخمگشایی نیز در شدت 1000 لوکس و در 18 ساعت روشنایی به میزان 1/7±08/83 درصد و کمترین آن 7/2± 17 درصد و در تیمار تاریکی مطلق تعیین شد. در تیمارهای مختلف تخمهای این گونه قابلیت تخمگشایی را داشته، اما با کاهش مدت زمان حضور نور درصد تخمگشایی کاهش یافته است. مدت زمان رسیدگی جنسی، طیکردن مراحل ناپلیوسی و کوپهپودیت در شدت 50 لوکس و در 24 ساعت روشنایی سریعتر بوده و مدت آن 2/1±5/9 روز تعیین شد. همچنین نسبت جنسی بالغان در تیمارهای مختلف اختلاف معنیدار آماری نداشتهاند. بنابراین برای تولید تخم و درصد تخمگشایی بالاتر و زمان بلوغ کوتاهتر، بهترین شرایط شدت نور پایین و دورة نوری 18 ساعت تعیین شد و نور ممتد با شدت بالا و تاریکی مطلق نیز برای این گونه نامناسب تشخیص داده شده است.

تأثیر سطوح مختلف عصارة زنجبیل بر کارایی رشد، تغذیه و ترکیبات بیوشیمیایی لاشة ماهی بنی (Mesopotamichthys sharpeyi) انگشتقد
مطالعة حاضر با هدف تعیین تأثیر سطوح مختلف عصارة زنجبیل بر کارایی رشد، تغذیه و ترکیبات بیوشیمیایی لاشة ماهی بنی در مرحلة انگشتقدی به مدت 8 هفته انجام شد. تعداد 192 قطعه بچهماهی بنی با متوسط وزن اولیة 32/0±67/11 گرم در قالب طرحی کاملاً تصادفی به 4 تیمار با 3 تکرار برای هر تیمار تقسیم شدند. ماهیها روزانه به میزان 3 درصد وزن بدن و در 3 نوبت با جیرة غذایی حاوی عصارة زنجبیل با سطوح 0، 1/0، 5/0 و 1 درصد (به ترتیب بهمنزلة تیمار کنترل و تیمارهای 1، 2 و 3) تغذیه شدند. گروه فاقد عصارة زنجبیل بهمنزلة تیمار کنترل در نظر گرفته شد. بیشترین میزان شاخص افزایش وزن بدن و شاخص کبدی در بین تیمارهای آزمایش در تیمار 3، تغذیهشده با جیرة حاوی 1 درصد عصارة زنجبیل، مشاهده شد (P<0.05). مطلوبترین میزان ضریب تبدیل غذایی و نرخ رشد ویژه مربوط به تیمار 3، تغذیهشده با جیرة حاوی 1 درصد عصارة زنجبیل، بود (P<0.05). میزان ضریب چاقی و ضریب بازده پروتئین بین تیمارهای مختلف اختلاف معنیداری نشان ندادند (P>0.05). نتایج نشان داد که استفاده از عصارة زنجبیل در جیرة غذایی تأثیری در میزان پروتئین، چربی، خاکستر و رطوبت لاشة ماهی بنی ندارد (P>0.05). با این حال، بیشترین میزان پروتئین و چربی لاشه به ترتیب در تیمار 1 تغذیهشده با جیرة حاوی 1/0 درصد و تیمار 2 تغذیهشده با جیرة حاوی 5/0 درصد عصارة زنجبیل مشاهده شد. به طور کلی، میتوان نتیجه گرفت که عصارة زنجبیل موجب تحریک رشد شده، اما تأثیری در میزان ترکیبات لاشه نداشته است. بنابراین، میتوان استفاده از عصارة زنجبیل را بهمنزلة محرک رشد در ماهی بنی در مرحلة انگشتقدی پیشنهاد کرد.

بررسی شاخص مطلوبیت زیستگاه سیاهماهی (Capoeta gracilis, Keyserling 1861) در رودخانة طالقان
در اجرای طرحهای حفاظت از اکوسیستمهای آبی و ماهیان، وجود دانش کافی دربارة نیازهای زیستگاهی گونههای آبزی اهمیت بسزایی دارد. از این رو، این مطالعه با هدف شناخت شاخصهای مطلوبیت ویژگیهای زیستگاهی گونة سیاهماهی (Capoeta gracilis) در محدودة پراکنش آن در رودخانة طالقان انجام شد. برای بررسی ویژگیهای زیستگاهی این گونه در این رودخانه فراوانی این ماهی و متغیرهای زیستگاهی شامل ارتفاع، عمق، عرض، سرعت جریان، قطر متوسط سنگ بستر و دمای آب در 33 ایستگاه از پاییندست به سمت بالادست رودخانة طالقان در مهرماه 1392 بررسی شد و محدودة زیستگاه انتخابی این گونه به دست آمد. مطابق نتایج، شاخصهای مطلوبیت ویژگیهای زیستگاهی این گونه ارتفاع 1550-1400 متر، عمق در دامنة 55-40 سانتیمتر، عرض رودخانه با کمتر از 5 متر، سرعت در دامنة 6/0-3/0 متر بر ثانیه، قطر متوسط سنگ بستر در دامنة 45-30 سانتیمتر و دما در دامنة 18-16 درجة سانتیگراد قرار داشت. وجود این شرایط با شاخص مطلوبیت زیستگاه 813/0 نشان میدهد که رودخانة طالقان برای گونة سیاهماهی زیستگاهی با کیفیت بسیار عالی است.

بررسی اثر درجۀ حرارت در غلظت کشندة (LC50 96-h) سم سایپرمترین (Cypermethrin) در ماهی سفید دریای خزر (Rutilus frisii kutum, Kamensky, 1901)
طی این تحقیق سمیت حاد[1] حشرهکش سایپرمترین در سه سطح دمایی مختلف (15 و 20 و 25 درجۀ سانتیگراد) در بچهماهیان 1±3 گرمی (میانگین± SD) سفیدماهی دریای خزر (Rutilus frisii kutum Kamensky, 1901) به منظور تعیین غلظت کشندة 50 درصد از جمعیت ماهیان در 96 ساعت مطالعه و بررسی شد. آزمایشها به صورت ساکن[2] و بر اساس روش استانداردO.E.C.D به مدت 4 شبانه روز (96ساعت) انجام و پارامترهای مؤثر فیزیکوشیمیایی آب از جمله pH، سختی کل، اکسیژن محلول و درجۀ حرارت کنترل شد. آزمایشهای ابتدایی به منظور یافتن محدودة کشندگی با رهاسازی بچهماهیان به داخل آکواریومهایی که به حجم 20 لیتر آبگیری و به طور مداوم هوادهی میشدند به تعداد 10 عدد بچهماهی برای هر آکواریوم انجام شد و پس از بهدستآمدن محدودة کشندگی تیمارهای نهایی با 6 تیمار و 3 تکرار مشخص شد و در نهایت بر اساس نتایج و با استفاده از آنالیز پروبیت، مقادیرLC10 ، LC30، LC50، LC70، LC90 و LC99 در 24، 48، 72 و 96 ساعت برای هر سه سطح دمایی بر روی سفیدماهی اندازهگیری شد و سمیت حاد (LC50 96-h) سم سایپرمترین برای بچهماهی سفید در دماهای ºC 1±15، ºC1±20 و ºC 1±25 به ترتیب برابر54/1، 49/1 و 30/1 میکروگرم در لیتر به دست آمد. نتایج نشان داد با افزایش درجۀ حرارت سمیت سایپرمترین افزایش مییابد و مرگومیر بیشتری برای ماهیان اتفاق میافتد. در این آزمایش از نظر بالینی علائمی مانند خونمردگی در ناحیة ساقة دمی، تنفس غیرعادی، شنای نامنظم در ماهیان مورد آزمایش مشاهده شد.

بررسی خواص ضدباکتریایی عصارههای ریزجلبک Chlorella vulgaris جداشده از خلیج چابهار
میکروجلبکها منابع زیستی مهمیاند که طیف گستردهای از برنامههای کاربردی زیستفنّاوری را به خود اختصاص دادهاند. در این پژوهش تأثیرات ضدباکتریایی عصارههای آلی ریزجلبک کلرلا ولگاریس خالصسازیشده از خلیج چابهار علیه سه سویة باکتری گرم منفی اشرشیاکلای، ویبریو کلرا و پروتئوس ولگاریس و دو سویة باکتری گرم مثبت استافیلوکوکوس اورئوس و لیستریا مونوسیتوژنوس آزمایش شد. عصارههای اتانولی و کلروفورمی بهدستآمده به دو روش انتشار دیسک و رقتهای متوالی در لوله به منظور تعیین حداقل غلظت بازدارنده (MIC) و حداقل غلظت کشندگی (MBC) بررسی شدند. بر اساس نتایج، عصارة اتانولی بیشترین اثر را در باکتری ویبریو کلرا نشان داده است و حداقل غلظت کشندگی برای باکتری اشرشیاکلای به دست آمد. عصارة کلروفورمی بر باکتری پروتئوس ولگاریس بیشترین اثر را نشان داد و با غلظتهای متفاوت حداقل غلظت کشندگی برای همة باکتریهای مورد آزمایش به دست آمد. به طور کلی، ریزجلبک کلرلا ولگاریس بومی ایران دارای اثر ضدباکتری قوی است و عصارة کلروفورمی نسبت به عصارة اتانولی دارای اثر بیشتری در باکتریها بود.

بررسی اثر عصارة آویشن (Zataria multiflora) در کیفیت شیمیایی سوریمی تولیدشده از ماهی کپور معمولی (Cyprinus carpio) طی نگهداری در یخچال
طی مدت 15 روز نگهداری (1، 3، 6، 9، 12 و 15) در یخچال (1±4 درجة سانتیگراد) بررسی شد. طی دورة نگهداری به فاصلة زمانی هر سه روز یکبار آزمونهای شیمیایی از قبیل تیوباربیتوریک اسید (TBA)، مجموع بازهای نیتروژنی فرار (TVB-N) و pH روی نمونهها در سه تکرار انجام شد. نتایج نشان داد که میزان تیوباربیتوریک اسید در روزهای پایانی در نمونة شاهد به طور معنیداری بیش از نمونههای تیمارشده با عصارة آویشن بود. مجموع بازهای نیتروژنی فرار در نمونة شاهد از روز سوم تا پایان دوره از حد مجاز بالاتر بود و در نمونههای تیمارشده با عصارة 2 درصد آویشن در روز آخر به 2/39 میلیگرم نیترژن در 100 گرم گوشت رسید، ولی در نمونههای تیمارشده با عصارة 4 درصد آویشن تا روز آخر نگهداری قابل قبول بود. مقدار pH در تمام طول دورة نگهداری در نمونههای شاهد به طور معنیداری بیش از نمونههای تیمارشده با عصاره بود (05/0>p). بر اساس یافتههای پژوهش حاضر مشخص شد که عصارة 4 درصد آویشن میتواند به منظور افزایش ماندگاری سوریمی در شرایط نگهداری کوتاهمدت یخچال استفاده شود.

این مطالعه با هدف ارزیابی تأثیرات فلوکستین در رفتارهای خشونتی جنسیت نر در مقابل ماده و غلظت هورمون تستوسترون آن در ماهی جنگجو (Betta splendens) طی یک دورة تولیدمثل انجام شد. ماهیان نر به مدت شش روز در معرض غلظتهای صفر (گروه کنترل) و µg/L 54/0 (گروه در معرض قرار داده شده) فلوکستین قرار داده شدند و رفتارهای خشونتی ماهی نر در مقابل ماده «مزاحم» در مراحل قبل از لانهسازی، بعد از ساخت لانه، بعد از تخمریزی و بعد از تفریخ تخمها اندازهگیری شد. پس از 6 روز، سطوح تستوسترون در گروه در معرض قرار داده شده نسبت به گروه کنترل کاهش معنادار نشان داد. رفتارهای خشونتی در اغلب مراحل تولیدمثلی از فلوکستین تأثیر گرفت و تفاوت تعداد رفتار رفت و برگشت به لانه در مراحل بعد از لانهسازی و بعد از تخمریزی نیز بین دو گروه از لحاظ آماری معنادار بود. نتایج این مطالعه مشخص میکند که فلوکستین در غلظت کم (µg/L 54/0) با تأثیر در سیستم درونریز ماهی جنگجو موجب کاهش سطوح کلی تستوسترون میشود و خشونت جنسیت نر را در تقابلات مستقیم با ماده طی تولیدمثل کاهش میدهد.

تنوع ژنتیکی اردکماهی (Linnaeus, 1758; Esox lucius) در شرق دریای خزر با استفاده از نشانگر ریزماهواره
اردکماهی یکی از گونههای اقتصادی و باارزش حوزة جنوبی دریای خزر است که اطلاعات دربارة جمعیتهای مختلف آن در حوزة جنوبی خزر در دسترس نیست. با توجه به اهمیت شناخت تنوع ژنتیکی در مدیریت ذخایر و حفاظت از گونهها در این مطالعه از هفت جفت آغازگر ریزماهواهای برای بررسی 90 نمونه اردکماهی از سه منطقة دهانة رودخانة تجن، آببندانهای ایزدشهر و تالاب لپوی زاغمرز واقع در استان مازندران استفاده شد. میانگین تعداد آلل مشاهدهشده در نمونههای رودخانة تجن 286/0 و ایزدشهر 010/1 و تالاب لپوی 800/0 و میانگین هتروزیگوسیتی مشاهدهشده و مورد انتظار برای مجموع نمونهها به ترتیب 383/0 و 621/0 محاسبه شد. نتایج آزمون انحراف از تعادل هاردی- واینبرگ نشان داد که تنها یکی از نمونهها در تعادل قرار داشت. با توجه به نتایج، علائمی از تنگنای ژنتیکی در جمعیتها دیده شد. دندروگرام ترسیمی و آزمون واریانس مولکولی بیانگر وجود سه جمعیت مجزا در سه منطقة مورد مطالعه بود که میبایست در مدیریت ذخایر این گونه مورد توجه قرار گیرد.

رشد و تغییرات بیوشیمی بدن بچهماهی سفید دریای خزر (Rutilus frissii Kutum) تحت تأثیر سطوح مختلف پروتئین و چربی جیره
این تحقیق به منظور بررسی تأثیرات سطوح مختلف پروتئین (40، 35 و 30 درصد) و چربی (14، 12 و 10 درصد) در عملکرد رشد و تغییرات فیزیولوژیک بچهماهی سفید دریای خزر (Rutilus frisii kutum) انجام شد. 675 ماهی (01/0 ± 15/1 گرم) در 27 آکواریوم 45 لیتری توزیع و به مدت 8 هفته در 4 وعدة غذایی غذادهی شدند. تقابل بین پروتئین و چربی تفاوت معنیداری در هیچیک از عوامل رشد ایجاد نکرد (05/0> P). نتایج نشان داد بهکارگیری پروتئین تا سطح 35 درصد موجب افزایش برخی از شاخصهای رشد از جمله وزن نهایی و افزایش وزن شده است (05/0 >P ). بهترین عملکرد رشد در ماهیان تغذیهشده با جیرههای حاوی 14 درصد چربی مشاهده شد (05/0 >P). بازده پروتئین و گلوکز تحت تأثیر تقابل بین پروتئین و چربی جیره قرار نگرفتند (05/0> P). میزان تریگلیسرید و کلسترول به طور معنیداری تحت تأثیر پروتئین و چربی و تقابل بین این دو قرار گرفت (05/0 >P )، بالاترین میزان تریگلیسرید در ماهیان تغذیهشده با 30 درصد پروتئین و 10 و 12 درصد چربی و بیشترین میزان کلسترول در ماهیان تغذیهشده با 40 درصد پروتئین به همراه 12 و 14 درصد چربی مشاهده شد. نتایج مطالعة حاضر نشان میدهد که پروتئین کمتر (30 درصد) و بیشتر (40 درصد) از حد نیاز ماهی سفید میتواند تأثیرات منفی در رشد و پارامترهای فیزیولوژیک داشته باشد و افزایش سطح چربی تا 14 درصد اثر منفی در پارامترهای رشد بچهماهیان سفید نداشت. به منظور عملکرد بهتر ماهی سفید در این محدودة سنی جیرهای با 35 درصد پروتئین و 14 درصد چربی پیشنهاد میشود.
