استخراج فایکوسیانین ازجلبک Arthrospira platensis با استفاده از حلال‌ها و روش‌های مختلف

چکیده

در این مطالعه، استخراج رنگدانه فایکوسیانین از میکروجلبک Arthrospira platensis با استفاده از حلال­ها (آب مقطر، بافر سدیم فسفات) و روش­های مختلف (سونیکیشن، مایکروویو و انجماد و ذوب) برای مقایسه و تعیین بهترین حلال و روش به منظور رسیدن به بیش­ترین بازده انجام شد. میزان 1/0 گرم پودر جلبک A. platensis در 10 میلی لیترحلال (آب مقطر و بافر سدیم فسفات (1/0 ± 7.4 pH=)( در سه روش استخراج سونیکیشن با دامنه 50درصد به مدت 15 دقیقه، مایکروویو در 100 وات به مدت 3 دقیقه و انجماد در 2 ± 20- درجه سانتی گراد به مدت 24 ساعت و ذوب در دمای اتاق انجام شد. تجزیه واریانس برای میانگین داده­ها± SD (انحراف معیار) با استفاده ازSPSS  در سطح اطمینان (05/0 P <) به دست آمد. بیش­ترین بازده استخراج توسط بافر سدیم فسفات و روش انجماد و ذوب (mg/g 55/1 ± 3/146PC = ) و کم­ترین توسط آب مقطر و روش مایکروویو ( mg/g68/2 ± 4/94 PC =) به دست آمد.

چکیده [English]

In this study, phycocyanin pigment extraction from Arthrospira platensis microalgae was carried out using various solvents (distilled water, phosphate buffered saline) and methods (sonication, microwave and freezing & thawing) for comparison and determine the best solvent and method in order to achieve higher extraction yield. Rate of 0.1 g A. platensis powder in 10 cc of solvent (distilled water and phosphate buffered saline) was similar in each three extraction method whereas the conditions of extraction methods sonication, microwave and freezing & thawing as follows, respectively: 50% amplitude and 15 min, 100 W and 3 min, frozen at -20 ± 2 °C for 24 h and thawing at room temperature. The analysis of variance was obtained for data mean ± SD (standard deviation) using SPSS in confidence level of (P < 0.05). Most extraction yield was obtained by phosphate buffered saline (PBS) and method Freezing & Thawing (pc= 146/3 ± 1/55 mg/g) and least by distilled water and method Microwave (pc= 94/4 ± 2/68 mg/g).
