ارزیابی عملکرد فیلتر ضدباکتریایی Zeolite-PEIدر مخازن پرورش پست لارو میگوی بزرگ آب شیرین (Macrobrachium rosenbergii‬)

چکیده

استفاده از موادی مؤثر به­ منظور مهار باکتری­ ها در محیط آبی به‌دلیل استفادة بی ­رویه و طولانی مدت از مواد ضدباکتریایی مانند آنتی ­بیوتیک ­ها ضروری به‌نظر می­ رسد. پلی‌اتیلن‌ایمین (PEI) یک پلیمر مصنوعی با خاصیت ضدباکتریایی و با فرمول شیمیایی n(C2H5N) شامل تعداد زیادی گروه­ های جانبی NH2 است و استفاده از این پلیمر در سطوح مختلف به ­منظور استفاده از خاصیت آنتی­ باکتریایی می­ تواند مورد نظر ­باشد: از جمله این سطوح که کاربرد زیادی در آبزی­ پروری دارد، زئولیت است. در این پژوهش، زئولیت­ با دو غلظت 1 و 2 میلی ­گرم در لیتر پلیمر PEI پوشش داده شد و پس از تأیید به‌وسیلة آنالیز طیف‌سنجی تبدیل فوریه مادون قرمز (FTIR)، درون فیلترها قرار داده شد و با سنجش بار باکتریایی آب مخازن و همچنین تأثیر بر بازماندگی و رشد پست ­لاروها، عملکرد فیلترها مشخص شد. نتایج نشان داد که کنترل منفی، تیمار زئولیت اصلاح‌شده با غلظت­ I، تیمار زئولیت اصلاح‌شده با غلظت­ II پلیمر PEI، تیمار زئولیت طبیعی و کنترل مثبت در بار باکتریایی CFU/ml 104 باکتری آئروموناس‌هیدروفیلا و در روز دهم آزمایش به‌ترتیب 50، 125، 125، 348 و 380 colony/ ml 100 در محیط کشت TSI رشد کرد که نشان ­دهندة کاهش 67/1 درصد بار باکتریایی در هر دو تیمار اصلاح شده با غلظت I و II پلیمر PEI نسبت به کنترل مثبت است. همچنین میزان بقا پست ­لاروها در تیمار زئولیت اصلاح شده با غلظت­ I و II نسبت به کنترل منفی افزایش معنی­ داری نشان داد (0/05>P). هر چند در سنجش وزن خشک و طول پست ­لاروها هیچ اختلاف معنی ­داری بین تیمارها دیده نشد (0/05<P). در نهایت پس از جمع ­بندی و مقایسة نتایج این پژوهش با سایر پژوهش ­ها می­ توان نتیجه گرفت که استفاده از زئولیت پوشش داده شده با پلیمر PEI در سیستم فیلتراسیون مخازن پرورش پست لارو میگوی بزرگ آب شیرین تأثیر بالقوه ­ای در کاهش بارباکتریایی باکتری آئروموناس‌هیدروفیلا دارد.

چکیده [English]

Use of effective substances to inhibit bacteria in the aquatic environment due to indiscriminate and long-term use of antibacterial substances such as antibiotics seems to be necessary. Polyethyleneimine (PEI) is a synthetic polymer with antibacterial properties and chemical formula (C2H5N)n which consists of many side groups of NH2, the use of this polymer due to its antibacterial property in binding with different substrates can be considered. Among these substrates that are widely used in aquaculture is Zeolite. In this study, zeolite was coated with PEI polymer with two concentrations of 1 and 2 mg/l and after confirmation by Fourier transform infrared spectroscopy (FTIR) analysis is placed inside the filters. The function of the filters was determined by measuring the bacterial load of the water, survival and growth of post larvae. The results showed that negative control, treatment of zeolite modified with concentration I, treatment of zeolite modified with concentration II of PEI polymer, treatment of natural zeolite and positive control at 104 CFU/ml Aeromonas hydrophila bacteria on the 10 days of the experiment were grew 50, 125, 125, 348 and 380 colony/100 ml in TSI medium, which shows a decrease of 67.1% of the bacterial load in both modified treatments with concentrations I and II of PEI polymer compared to the positive control. Also, the survival rate of postlarvae showed a significant increase in the treatment of modified zeolite with concentration I and II compared to the negative control (P<0.05). However, there was no significant difference between treatments in dry weight and length (P>0.05). Finally, after summarizing and comparing the results of this study with others, it can be concluded that the use of zeolite modified with PEI polymer in the filtration system of giant freshwater prawn Macrobrachium rosenbergii post larval rearing tanks has a potential effect on bacterial reduction of Aeromonas hydrophila bacteria.
